Insertion of a hygromycin resistance cassette into one allele of the Rb1 gene replaced part of intron 3 and introduced three nucleotide changes into exon 3, creating two termination codons and a new PstI site. This was accomplished in ES cells in which Rb1 had already been targeted. The resulting ES cells are Rb1tm1Tyj/Rb1tm2Tyj. (J:59268)
