A construct, in which exons 5 and 6 were flanked by a loxP site in intron 4 and a floxed neo cassette in intron 6, was incorporated at the endogenous locus via homologous recombination. The floxed exons encode amino acids 53 through 113, which encompass most of the Trp53-binding domain. (J:75118)
