A 3.3 kb full length human DSG3 cDNA under the control of the human keratin 1 promoter was used for the transgene. The promoter was used to target expression of the transgene to the upper (suprabasal) epidermal layer. Western blot analysis verified the presence of human protein in the epidermis of transgenic mice. Immunohistochemical analysis showed the presence of human protein in the suprabasal layers of transgenic epidermis. Southern blot determined that 13-15 copies of the transgene had integrated into the genome. (J:78071)
