A human CCKBR cDNA sequence driven by the rat elastase 1 promoter was used for the transgene. The -500 to +8 region of the rat elastase 1 promoter specifically targets expression of the transgene to pancreatic acinar cells. RT-PCR analysis of mRNA from pancreas, brain, and other tissues confirmed that the human gene is expressed only in the pancreas of transgenic animals. Western blot of pancreatic plasma membranes using antibodies directed against the third extracellular loop of human CCKBR demonstrated the presence of human protein in the pancreas of transgenic animals.
