A 2 kb region of the gene from exon 1 to exon 5 was replaced with a neomycin resistance cassette via homologous recombination. The gene targeting event deletes bp 107-718 of the cDNA sequence. Northern blot analysis of liver from homozygous mutant animals using a full-length cDNA probe did not detect gene expression. Polyclonal antibodies directed against the extracellular domain of the protein did not react with mouse embryonic fibroblasts derived from homozygous mutant animals thus confirming the null allele.
