Exons 2 and 3, encoding the amino terminal activation domains (residues 4 through 557), were replaced with a cassette containing IRES-lacZ and neo genes. Northern blot analysis of total RNA obtained from brain and heart tissue showed an absence of transcript in homozygous mutant mice. A lack of encoded protein was determined by Western blot analysis using an antibody directed against the carboxy terminal. Expression of beta-galactosidase was controlled by the promoter of the endogenous gene. (J:77894)
