The majority of exons 5-8 were replaced with a neomycin resistance gene in the opposite orientation via homologous recombination. The gene targeting event deletes 55 amino acids in the extracellular/juxtamembrane region, the transmembrane region, and the entire cytoplasmic tail of the encoded protein. Exons 5, 7, and 8 encode highly conserved domains across species. Absence of gene expression was demonstrated by Northern blot analysis of E16 fetal lung using a 5' region of the gene as a probe. Lack of gene expression was further verified by cell surface immunofluorescence analysis of E15 fetal liver cells using a monoclonal antibody to the mouse protein. (J:70294)
