Exon 2, which encodes the glutamine-rich activation domain, was replaced with a cassette containing an IRES-lacZ-neo fusion gene, adjoined to a splice acceptor site. The three isoforms produced from the endogenous allele were undetected in homozygous mutant mice by Western blot analysis of nuclear extracts. (J:60658)
