A neomycin selection cassette was inserted into exon 2 immediately downstream of the translational start codon. The disrupted exon encoded the secretion signal. Northern blot analysis showed an absence of transcript in the mammary glands of homozygous mutant mice. The encoded protein was not detected by Wester blot analysis of milk obtained from homozygous mutant mice. (J:18947)
