One loxP site was inserted in intron 3 and a floxed hygromycin selection cassette was inserted in intron 5. The hygromycin selection cassette was removed in ES cells by transient Cre expression, leaving exons 4 and 5 flanked by loxP sites in the final allele. (J:69471)
