A construct containing a promoterless lacZ gene adjoined to a neomycin selection gene was inserted 50 bp upstream of the translational start codon. The insertion introduced a frameshift mutation upstream of the DNA-binding domain. An absence of encoded mRNA and protein was determined by in situ hybridization and immunocytochemical analyses of homozygous mutant mice. (J:45064)
