3.4 kb of the gene encoding amino acids 27-301 was deleted and replaced with a PGK-neo cassette via homologous recombination. The absence of gene expression in homozygous mutants was confirmed by electrophoretic mobility shift assay (EMSA) and Western blot analysis of nuclear extracts from mutant mouse embryonic fibroblasts (MEF).
