A single loxP site was introduced upstream of exon 3 and a loxP site followed by an frt-flanked neomycin and thymidine kinase selection cassette was inserted 3' to exon 3. The selection cassette was removed in ES cells by transient FLPe recombinase expression prior to the production of chimeric mice, leaving loxP sites flanking exon 3 in the final allele. (J:75112)
