A single loxP site was inserted 5' to exon 10 and a loxP flanked neomycin and thymidine kinase cassette was inserted 3' to exon 10. Transient transfection of Cre recombinase in ES cells resulted in the deletion of the neomycin and thymidine kinase cassette, leaving single loxP sites flanking exon 10 in the final allele. (J:76211)
