A construct containing a beta-geo fusion marker as well as a splice-acceptor site and a picornaviral internal ribosome-entry site (IRES) was inserted upstream of exon 8 and 9. The construct was designed to prevent the translation of the functionally essential centromere targeting region. Southern blot analysis confirmed the targeting event. (J:45656)
