A neomycin selection cassette was inserted into the exon encoding the signal sequence, disrupting the reading frame downstream. Homozygosity in ES cells was achieved by gene conversion. FACS analysis of homozygous mutant ES cells showed an absence of protein surface expression. The absence of encoded protein mice derived from these clones was determined via immunohistochemical analysis. (J:30621)
