Part of exon 4 and all of exon 5, which encodes the S4 voltage sensor of domain I, were replaced with a PGK-neo cassette via homologous recombination. The PGK-neo cassette introduces stop codons in all 3 reading frames. RT-PCR analysis detected weak expression of mutant transcripts in homozygous mutant animals. (J:76456)
