Exons 4-8 encoding the pore-lining domain and neighboring transmembrane domains were replaced with a neo-URA3 cassette via homologous recombination. Western blot of brain extract from homozygous mutant animals confirmed the absence of protein product. Northern blot analysis also failed to detect gene expression. (J:66144)
