Disruption of the calpain small subunit gene was achieved by replacing the 3'-terminal half of exon 9 with a PGK-neo cassette. No calpain activity was detected in extracts from homozygous mutant ES cells. Although the large subunit genes were not altered and are transcribed, the resultant proteins are unstable in the absence of the small subunit. (J:74966)
