A neomycin resistance gene replaced most of exons 4 and 5 of this gene (encoding part of the hyaluronan binding domain and IM7 mAB recognition site) via homologous recombination. PCR and Southern Blot confirmed the integration of the neo gene in homozygous mutant animals. Synovial fibroblasts from homozygous mutant animals did not express mRNA as determined by Northern blot, and Western blot using IM7 mAB as a probe yielded negative results. (J:74268)
