A neomycin resistance cassette replaced exon 2 and flanking intronic sequences. RT-PCR analysis on RNA derived from homozygous embryos demonstrated that no detectable transcript was produced from this allele, and western blot analysis on liver lysates confirmed that no protein was expressed from this allele.
