A lacZ reporter was inserted in frame with the Hoxd12 gene, at the StyI site of the first exon. In addition, a loxP-PGKneo-loxP cassette was inserted next to the lacZ cassette, in the same transcriptional orientation. Excision of the selectable marker was conducted and the resultant clones were injected into blastocysts and transferred to the germ line. The resultant targeted allele had no deletion of genomic sequences. (J:36770)
