An HA tag followed by a stop codon was introduced into exon 8, and was followed by an IRES-lacZ gene and neomycin selection cassette that replaced exons 9, 10 and 11. Western blot analysis on homozygous embryos demonstrated that no detectable ENG protein was expressed from this allele; however, a beta-galactosidase protein was expressed under the control of the endogenous promoter.
