A floxed neomycin resistance cassette was inserted upstream of exon 3 and an additional loxP site was inserted downstream of the exon. Transient expression of Cre recombinase in ES cells prior to production of chimeric mice excised the neo cassette leaving exon 3 flanked by loxP sites. (J:73374)
