A 4.0 kb genomic fragment containing the coding sequences for amino acids 31 to 560 was replaced with a neomycin selection cassette. These sequences correspond to the tail and alpha coiled coil domains and half of the motor domain. Northern blot analysis on RNA derived from brain of homozygous mice confirmed that no detectable transcript was produced from this allele. In addition, western blot analysis demonstrated that no encoded protein was detectable in brain lysates derived from homozygous mice. (J:68003)
