A genomic fragment containing exon 1 was replaced with a neomycin selection cassette inserted in reverse transcriptional orientation to the gene. Northern blot analysis demonstrated that that no detectable transcript was present in kidney or liver tissue of homozygous mice, and western blot analysis confirmed that no detectable protein was present in embryonic fibroblasts derived from homozygous mice.
