A mouse cDNA containing the sequences encoding amino acids 14 to 304 of the Atp1b1 protein was inserted in frame into exon 1, at codon 18 of the Atp1b2 gene. A loxP flanked neomycin cassette was inserted 3' to the cDNA sequence. Northern blot analysis indicated that a mutant fusion transcript is produced from this allele, and western blot analysis on brain membrane fractions indicated that increased levels of Atp1b1 protein were detected, and expression of Atp1b2 was eliminated in homozygous mice.
