A PGK-neomycin resistance cassette replaced most of the first coding exon and 30bp of 3' flanking sequence, including the initiation codon and sequences encoding most of the MH1 domain. Northern analysis of 9.5dpc homozygous embryos detected a shorter transcript, but not the wild-type transcript, using a probe for the 3' UTR. Western blot analysis using an antibody that recognizes the MH2 domain did not detect intact or truncated protein in 9.5dpc homozygous embryos. (J:71998)
