LoxP sites were inserted flanking exon IIIb as follows: A loxP flanked neomycin reistance cassette was inserted in the intron 3' to exon IIIb and an additional loxP site was inserted into the intron 5' to exon IIIb. Correctly targeted ES cells were transiently transfected with a vector expressing Cre recombinase, and lines were selected that retained exon IIIb with flanking loxP sites, and used to inject blastocysts.
