A mutation was introduced to create a stop codon immediately upstream of the coding region of the CGRP alternative splice transcript in exon 5. The codons for a 4-amino-acid peptide (DLQA) of unknown function that is normally proteolytically spliced from the C-terminus of the alphaCGRP prohormone, was duplicated and inserted after the codons for a dibasic proteolytic cleavage site (normally used to cleave CGRP from the alphaCGRP prohormone) and before the stop codon in order to preserve the expression of this peptide. Thus, only the CGRP expression is knocked-out and the expression of the other neuropeptides created from this locus (calcitonin, etc.) is left undisturbed. (J:57708)
