A neomycin selection cassette replaced exon 1, which contains the translation start site and the sequences encoding the first 26 amino acids of the protein. RT-PCR analysis on RNA derived from homozygous embryos demonstrated that no detectable transcript is produced from this allele, and western blot analysis on embryo extracts derived from homozygous mice confirmed that no stable protein is made.
