The entire coding region (exon 2), with the exception of the initiator methionine, was replaced with a neomycin selection cassette. The cassette was inserted in the same transcriptional orientation as the gene in order to preserve the expression of an adjacent loci encoding coronin. Immunoblot analysis showed an absence of encoded protein in lysates derived from the thymi and spleens of homozygous mutant mice. Additional analysis showed that coronin protein levels were not affected by the targeting event. (J:58711)
