A lacZ-PGK-neo cassette was inserted via homologous recombination resulting in deletion of most of the gene product, including loss of all LXXLL motifs and nuclear receptor coactivator function. Reporter gene assays showed expression was greatly reduced in mouse embyonic fibroblasts cultured from homozygous mutant embyros. (J:61840)
