A cassette containing both a lacZ coding sequence and a neomycin resistance gene was used to replace exons 4 and 5 which code for the DNA binding domain. This insertion inactivates both the beta1 and beta2 isoforms. RT-PCR was used to demonstrate a lack of mRNA in homozygous mutant animals. (J:52829)
