The gene was disrupted by the introduction of a lacZ-neo cassette into the fourth exon via homologous recombination. Immunohistochemistry of acinar cells from pancreas of homozygous mutant animals did not detect protein product, and was positive for beta-galactosidase activity. RNase protection assay detected faint transcript (5% of wild-type signal) in adult homozygous mutant animals thought to be a chimeric transcript incorporating the lacZneo sequence. (J:51627)
