A neomycin cassette was inserted into the first exon and downstream from the initiating codons that encode the A and B isoforms to effectively disrupt transcription of both isoforms. Northern analysis indicated a lack of gene specific transcript. Competitive binding assays for the progesterone receptor indicated a lack a functional protein in homozygous mutant animals. (J:28980)
